A tyrosine to phenylalanine substitution at position 145 (Y145F) was generated by a targeted A to T point mutation within exon 7 of the gene locus. For targeting purposes, a loxP and FRT flanked neomycin resistance cassette was placed downstream of exon 7. The selection cassette was subsequently removed by transient cre expression, leaving behind single loxP and FRT sites. The presence of the point mutation did not affect gene expression as determined by western blot analysis of thymocytes. (J:132995)
