The neuron-specific promoter elements of the mouse Thy1 gene drive expression of the 383 amino acid isoform of human tau - the shortest of the isoforms having four microtubule binding repeats (0N4R) - in which proline is replaced by serine at amino acid position 301 of the protein (P301S). Immunoblot analysis demonstrates the presence of human tau at approximately twice the level of the combined endogenous mouse tau proteins in brains and spinal cords of 5-6 month old homozygous transgenic mice.
