Fertilized eggs homozygous for the null Rag2tm1Fwa allele were co-injected with transgenes encoding the rearranged Tcra and Tcrb genes isolated from the CD8 positive T cell clone N15. These transgenes encode a T cell receptor specific for the vesicular stomatitis virus nuclear octapeptide antigen (VSV8) presented by the MHC-I variant Kb. Expression of the transgenes by thymocytes was confirmed by flow cytometry analysis using an antibody specific for the transgenic TCR. (J:133112)
