A loxP-flanked neomycin selection cassette was inserted upstream of exon 1 with an additional loxP site inserted downstream of the exon. Transient expression of cre recombinase removed the neomycin selection cassette leaving exon 1 flanked by loxP sites. Correct targeted was confirmed through Southern blotting. (J:132855)
