A Lexicon gene trap vector was inserted into the first intron at nucleotide position 1085, which is 666 bp upstream of the ATG. The gene trap was integrated between the transcriptional start sites of two of the three alternative first exons, 1b and 1c. RT-PCR confirmed presence of 1a- and 1b-NEO transcripts and absence of wild-type 1a and 1b transcripts in homozygous E9.5 embryos. Although 1c-WT transcript was not trapped by the insertion, it was barely detectable in the homozygous. In situ hybridization and the western blot confirmed absence of all three full length transcripts and mature proteins. (J:132031)
