ES cells containing the Tcrbtm1Slek mutation that lacks the Dbeta2-Jbeta2 locus were again targeted to replace the Vbeta14 recombination signaling sequence (RSS) with germline Dbeta1 and Jbeta1.1 segments, their RSS, and intervening intronic sequence. A floxed neo resistance cassette used in the targeting was subsequently removed by transient Cre expression. The gene targeting also introduced a loxP site and an EcoRI site to distinguish between the targeted allele and the endogenous Vbeta 14 allele. Southern blots of thymus confirmed the presence of both germline and recombined versions of the knocked in gene segments. (J:131995)
