A floxed Puro cassette and a nucleotide substitution that resulted in an alanine for serine amino acid substitution at position 1121 (S1121A) were knocked into the open reading frame. The puromycin resistance cassette can be removed by germ line cre-mediated recombination to produce a conditional knock-in allele. (J:106531)
