This allele of the gene locus was constructed in two steps. A loxP site flanked neomycin resistance gene cassette was inserted upstream of the Trav6d-3 (Valpha6/Vdelta3) segment located at the boundary of the Trav and Trdv gene segments. After targeting, the neo cassette was removed by transiently expressing Cre recombinase to leave in its place a single loxP site. ES cells were then targeted downstream of the Tcrd locus to replace the Traj57 (Jalpha57) segment with a recombined Trav9-2/Jalpha57 segment. Upstream of the recombined element a neo cassette and a loxP site were inserted. ES cells were screened by Southern blotting for targeting events that occurred in cis. These mice have an intact Tcrd locus that is flanked by loxP sites and have a recombined Valpha-Jalpha segment downstream that partly encodes a T cell receptor that recognizes the HY male antigen.
