A neo cassette with a 5' loxP site was inserted into intron 3 and an additional loxP site was inserted into intron 2. Targeting took place in PC3 ES cells, which are homozygous for Tg(Prm-cre)70Og, a cre transgene active only in haploid spermatids. A minority of the alleles created from this construct exhibited a deficiency of cre-mediated recombination driven by the male-germline specific protamine promoter in male germ cells and transmitted the conditional allele through male chimeric mice.
