A polyA based transcriptional stop element was inserted 1.5-kb downstream of the transcription start and a self-excising selection cassette was placed downstream of the gene. After recombination and generation of chimeric mice, the selection cassette excised itself in the male germline leaving behind the stop element and a single loxP site. Heterozygous mice that inherit the deletion paternally transcribe a truncated RNA that is missing the distal 58.5 kb of sequence. (J:108700)
