A polyadenylation signal in the opposite orientation preceded by a floxed neo cassette was inserted downstream of the endogenous promoter. The neo cassette was removed by cre-mediated recombination. Expression upstream and downstream of the insertion was confirmed by RNase protection assay and RT-PCR. (J:130529)
