A polyadenylation signal in the correct orientation followed by a floxed neo cassette was inserted downstream of the endogenous promoter. The neo cassette was removed by cre-mediated recombination. The expression of RNA upstream of the insertion and the absence of expression of RNA downstream of the insertion was confirmed by RNase protection assay and RT-PCR. (J:130529)
