The single-codon W298A mutation was inserted into the endogenous Ptf1a locus of a mouse embryonic stem (ES) cell line by RMCE using Ptf1atm1Mgn. The exchange cassette contained the Ptf1a gene SalI-SalI fragment from strain 129 genomic DNA with codon 298 tryptophan (TGG) changed to alanine (GCG) by PCR mutagenesis. (J:125691)
