A human cDNA encoding the short isoform (1072 amino acids) of RET was mutated by PCR to introduce the RET MEN2B mutation (Met918Thr). This mutated cDNA was inserted downstream of a human DBH promoter (5.8 kb). Between the transcriptional start site and the mutated cDNA, the construct contains the first intron of the rat preproinsulin gene (+13 +183 nt). A polyadenylation site from the murine protamine 1 gene was inserted at the 3' end. This transgenic line has a copy number of 70. (J:108163)
