A transgene consisting of the coding sequence of Thy1a (Thy1.1, CD90.1) followed by SV40 poly A sequence was inserted into a mouse Il10 gene contained in a BAC. The reporter cassette was inserted one codon after the ATG start site and a premature stop codon was included after the poly A signal to prevent downstream expression of the Il10gene. The modified BAC, including 113 kb of genomic DNA upstream and 83kb genomic DNA downstream of the reporter cassette, was injected into single-cell embryos. One founder line containing 12 copies of the reporter transgene was selected for further study. Transgene expression was confirmed in T cells cultured under Th2 polarizing conditions. (J:125255)
