Insertion of a bicistronic reporter construct into exon 1 inactivated the gene and brought both lacZ and PLAP under control of the endogenous promoter. The construct contains a bacterial beta-galactosidase open reading frame followed by a retroviral internal ribosome entry site (IRES) element, a human PLAP open reading frame, and a floxed neomycin resistance gene (lacZ-IRES-PLAP-loxP-neo-loxP). In heterozygous mice, lacZ is expressed in the cerebral cortex and in the hippocampal formation of Ammon's horn. (J:128544)
