The exons 3-6 that encode the catalytic domain of the protein were replaced with a neomycin resistance cassette. Elimination of gene expression in homozygotes was confirmed by quantitative RT PCR on liver RNA and by the lack of enzymatic activity of the gene product in plasma. (J:125786)
