A 5527 bp fragment of the mouse Nphs1 gene was used as the transgene promoter. Two ATG sequences in the first intron of the fragment were disrupted by site-directed mutagenesis to prevent initiation of transcription of Nphs1. A fragment containing the human IL2RA (CD25) cDNA, FLAG sequence, and polyadenylation signal of the human growth hormone gene were placed downstream of the Nphs1 fragment in a transgenic construct. The vector was injected into (C57BL/6 x DBA/2) x C57BL/6N fertilized eggs. Two lines 9 and 18 showed essentially the same phenotype, and line 18 was maintained by backcrossing with C57BL/6N mice. Immunohistochemistry demonstrated that the transgene is expressed in podocytes, and RT-PCR showed transgene expression in the brain and pancreas but not in other organs (heart, spleen, thymus, lung, or muscle).
