A gene trap vector containing the beta-geo cassette was inserted into the intron between exons 6 and 7 creating a truncation. Insertion of the gene trap was confirmed by PCR and Southern blot analysis. Disruption in protein expression was confirmed by western blot. Maternal expression in peripheral tissues such as fat, muscle, liver, pancreas, heart, lung, kidney and spleen is disrupted by insertion of the gene trap. However, paternally inherited expression in the brain is unaffected. (J:125128)
