Exons 6 through 8 were replaced with a cDNA encoding exons 6 through 8 with a modification in exon 8 that creates a tyrosine to phenylalanine substitution at amino acid position 449. This substitution disrupts the YXXM motif that when phosphorylated binds to the Src homology 2 domain of PI3 kinase to activate downstream pathways. A floxed neo cassette inserted downstream of exon 8 was removed by cre-mediated recombination. Expression and subcellular localization, as determined by western blot and flow cytometry, are not altered. Downstream signaling through Erk and p38 MAPK are unaffected whereas signaling via STAT5 is affected. (J:125360)
