This transgene consists of an optimized reverse tetracycline transactivator under the control of the mouse alpha myosin heavy chain. rtTA is optimized by changing 5 amino acids (S12G, E19G, A56P, D148E, and H179R referred as M2) in the synthesized (S2) construct and linking it to 3 repeats of a 12-amino-acid sequence, FFF, which is the minimum activator domain of VP16.
