A fragment consisting composed of the sequence ot the terminal 100 amino acids of the beta-amyloid precursor protein APP gene (C100, referred to as C104 in J:105050 which states that the transgene contains the terminal 100 amino acids of APP) was inserted into a plasmid encoding the neural-specific dystrophin (DMD) promoter. The vector also contained the SV40 early splice and polyadenylation sequences. A hydrophilic 10-amino acid FLAG sequence was engineered onto the aminoterminus of the C100 fragment. The construct was injected into (C57BL/6 x SJL)F2 fertilized eggs. Six founder mice were identified, 5 showing higher expression than the identified line 2. Line 18 was chosen to represent the higher expressing transgenic lines. RT-PCR revealed expression of the FLAGC100 in brains of all transgenic lines. (J:100978)
