A transgene construct composed of the signal peptide and 99 residues of the carboxy-terminal fragment of human amyloid beta precursor protein (BAPP) under control of the cytomegalovirus enhancer/chicken beta-actin (ACTB) promoter was injected into fertilized (C57BL/6 x C3H)F1 x C57BL/6N hybrid mouse oocytes. Transgenic mRNA was detected in many tissues. Amyloid beta was detected biochemically in brain, kidney, and to the largest extent in the pancreas. Marked accumulation of human amyloid beta peptides 30-fold higher than in normal human plasma) was found in plasma of transgenic mice. (J:100977)
