A neomycin resistance gene replaced 55 nucleotides in exon 2. The insertion was predicted to result in correct translation of the initial 47 amino acids followed by 100 random amino acids and a stop codon. RT-PCR confirmed correct integration of the mutation. An aberrantly spliced transcript was detected, where exon 1 was fused to exon 3, however, the production of protein from this transcript is unlikely. (J:123742)
