The K14-rtTA transgene contains the 2.3 kb fragment of the human keratin 14 promoter that drives expression of the reverse tetracycline transactivator gene (rtTA), followed by an SV40 polyadenylation sequence. The 4.9 kb transgene was injected into one-cell (C57BL/6 x SJL) mouse zygotes. Mice from founder line 208 found to harbor the transgene on the X chromosome (also called founder line K14-X). Random X inactivation may result in mosaic rtTA expression patterns in female mice. The transgene when combined with mice carrying a gene under the control of a tetracycline-responsive promoter allows doxycycline-inducible gene expression in skin cells. (J:55970, J:96886)
