A loxP site flanked Gfra1 cDNA (with SV40 poly(A) signal sequences), a GFP gene (with poly(A) signal sequences), and an FRT site flanked neomycin resistance gene cassette were introduced into exon 2. 95 Nucleotides containing the 5' UTR, the initiator ATG codon and the signal sequences, were deleted. Expression of the endogenous gene was abolished, however, the inserted cDNA (but not the GFP gene) was expressed. The neo cassette was removed through subsequent flp-mediated recombination.
