The transgene contains a mouse anti-Ctla4Ig/Cd80 fusion cDNA from which a cell surface bound form of the CD152 antigen (Ctla4Ig) is expressed under control of the IgM heavy chain promoter and enhancer, which directs expression to B cells. To prevent reverse signaling in the transgene expressing cell, the construct was created using a DNA fragment expressing the transmembrane domain from Cd80(B7-1), which lacks the C-terminal, intra-cytoplasmic tail.
