A bacterial artificial chromosome (BAC) was modified by inserting a humanized Renilla GFP (hrGFP, Stratagene) sequence in-frame with the ATG transcriptional start site of the mouse pro-opiomelanocortin-alpha (Pomc1) gene (this also deleted the first 30 nucleotides of Pomc1). Correctly transformed and targeted bacteria were used to isolate the 150 kb BAC, which was then injected into the pronucleus of fertilized one-cell stage FVB embryos. Transgenic line 1 was subsequently established. When exposed to UV light, brain tissue shows GFP fluorescence patterns consistent with endogenous Pomc1 expression. (J:120992, J:125165)
