EGFP cDNA followed by a stop codon was inserted in-frame after codon 396 in exon 11 of Foxp3 and a loxP-flanked PGK-Neo was inserted in exon 9 in the same orientation of Foxp3. A diphtheria toxin cassette for negative selection was included in the targeting construct. Correctly targeted chimeric males were crossed with Cre-deleter mice on the BALB/c background to remove the PGK-cassette. Mice express a mutant Foxp3/EGFP protein with the 33 amino acid C-terminal peptide residues including DNA- and transcription factor-binding and a nuclear localization sequence replaced by EGFP under control of the endogenous promoter (J:120636)
