Constructs were created, with the cDNAs for both the alpha chain (Valpha4) and the beta chain (Vbeta11) of the T cell receptor from the mouse CTL F5 clone, inserted in the 5' UTR of the human CD2 minigene carrying a frameshift mutation in its coding region. The CD2 minigene provides an intron, the 3' LCR and a polyadenylation signal which are sufficient to direct expression of the transgenes. The constructs were coinjected into C57BL/10 oocytes. Transmission of the transgenes produced several lines, and Southern blot verified multiple copies of the alpha and beta chain genes in one line.
