Male pronuclei of fertilized oocytes were microinjected with a transgenic construct comprising the human apolipoprotein C-III gene, including 2.5 kb of 5' and 1.1 kb of 3' flanking sequence. 1-2 copies of the transgene were estimated present per haploid genome by quantitative Southern blot analysis. Solution hybridization RNase protection analysis with a riboprobe representing the 3' untranslated region of human APOC3 demonstrated transgene-derived RNA in liver and intestine, but none in brain of transgenic mice. Electroimmunoanalysis with a human-specific anti-APOC3 antibody detected human APOC3 in plasma of mice hemizygous for the transgene at 3.15 +/- 0.3 mg/dl mice and in plasma of homozygous transgenic mice at 8.2 +/- 1.0 mg/dl.
