Male pronuclei of fertilized oocytes were microinjected with a transgenic construct comprising the human apolipoprotein C-III gene, including 2.5 kb of 5' and 1.1 kb of 3' flanking sequence. 100-110 copies of the transgene were estimated present per haploid genome by quantitative Southern blot analysis. Solution hybridization RNase protection analysis with a riboprobe representing the 3' untranslated region of human APOC3 demonstrated high levels of transgene-derived RNA in liver and lower levels in intestine, but none in brain of transgenic mice. Electroimmunoanalysis with a human-specific anti-APOC3 antibody detected human APOC3 at 54 +/- 11 mg/dl in plasma of transgenic mice. (J:81184)
