A retroviral vector was inserted into the third intron and contained a splice acceptor, termination codons in all reading frames, a selection marker and a transcription terminator. This design would interrupt the transcript before exon 4, which contains TM7 of the 7-transmembrane domain and the C-terminal domain. RT-PCR confirmed absence of wild-type transcript in mutants.
