A floxed neo was cassette inserted into intro 3 and an additional loxP site was placed in the 5' region of the gene via homologous recombination. All of the floxed sequence was then removed by transient Cre expression in the ES cells. Western blot analysis confirmed absence of protein expression in whole eye lysates from homozygous mice.
