A mini-gene encoding a codon-optimized cre, the SV40 polyadenylation signal and an Frt-flanked neo selection cassette was inserted into the first exon of the Ptcra (pTalpha) gene in ES cells. The neo was excised in vitro by transient transfection with flippase. Expression of cre is observed in immature DP thymocytes and in more mature SP cells. (J:117041, J:197923)
