The recombined immunoglobulin genes present in the HyHEL10 hybridoma produce antibodies with high affinity for hen egg lysozyme (HEL). The recombined murine Igk variable and joining gene segments present in this hybridoma were placed under the control of a Vkappa22-33 promoter. A floxed neomycin resistance cassette was placed upstream and the entire construct replaced the endogenous Igk-J1 gene segment. Mice were crossed with cre-transgenic mice to removed the floxed neomycin cassette. The transgenic IgK light chain is detectable on some B cells in heterozygote mice. (J:115079)
