A construct encoding a fusion protein comprising the mitochondrial targeting sequence from human cytochrome c oxidase subunit 8A (ubiquitous) joined to an enhanced yellow fluorescent protein (EYFP) was placed downstream of the tetracycline responsive element (tetO) and upstream of a beta-globin polyadenylation signal. A mouse with this transgene can be bred with another transgenic mouse expressing reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) under control of a selected promoter to produce bitransgenic mice in which mitochondrial EYFP expression can be regulated in a tissue-specific manner by administration/removal of the tetracycline analog, doxycycline.
