This transgene produces a fusion protein consisting of three cytotoxic T lymphocyte (CTL) epitopes and enhanced green fluorescent protein under control of the ubiquitously expressing chicken beta-actin promoter. The CTL epitopes are: 1) amino acids 1-80 of the lymphocytic choriomeningitis virus (LCMV) glycoprotein, which include the immunodominant H2-Db-restricted CTL epitope GP33-41; the H2-Db-restricted LCMV nucleoprotein-derived epitope NP396-404; and the H2-Kb-restricted E. coli beta-galactosidase-derived epitope betaGal497-504. Cre recombinase-mediated removal of a loxP-flanked STOP cassette between the promoter and the coding sequence activates the gene. Although CD11c+ dendritic cells bearing this transgene and expressing Cre recombinase under control of the Itgax (Cd11c) promoter do not fluoresce, immunohistochemical analysis demonstrates that approximately 8% of them present the LCMV nucleoprotein epitope.
