A replacement vector was used to insert a cassette containing: an SpeI linker with stop codons in all three reading frames, a retroviral internal ribosomal entry site (IRES) element fused to the lacZ reporter gene with a 3' polyA signal, and the hygromycin gene into a site in exon 3 of the Il5 gene.
