A targeting vector inserted GFP coding sequences and an frt-flanked hygromycin resistance gene into exon 1. Transient transfection with a vector encoding flp recombinase removed the hygromycin resistance gene and one of the frt sites from targeted ES cells. The expression pattern of GFP in homozygous mutant embryos and adult mice mirrors that of the endogenous gene. (J:114151)
