A 2.3 kb fragment of the Valpha gene, containing 2 kb of 5' flanking sequence was fused to a 195 bp cDNA fragment covering the unique V-J junction, with the remaining nucleotides of the Jalpha region plus ~100 bp of JC intron sequence. This VJ segment was inserted upstream of a 9.8 kb Calpha clone containing all the Calpha exons and extending .5 kb downstream of the polyA signal. A 683-bp fragment containing the immunoglobulin heavy-chain enhancer (EH) was inserted at the junction between the VJ and Calpha segments. (J:69570)
