A sequence encoding a floxed GFP-FRT-puro cassette followed by a form of Pax3 that acts as a transcriptional repressor was targeted to the locus. The repressor sequence contains the N-terminus of the Pax3 DNA-binding domains fused to the repression domain of the Drosophila Engrailed transcriptional repressor. Cre mediated recomibination results in the loss of the floxed GFP-FRT-Puro cassette and creates a dominant negative allele. (J:112275)
