LoxP sites were introduced into introns 7 and 9 and and frt-flanked neomycin resistance cassette was inserted within the loxP-flanked region in intron 9. The neomycin resistance cassette was removed by crossing to Flp- expressing mice, leaving only the loxP sites that flank sequences corresponding to the DNA-binding region of the encoded protein.
