A single loxP site was inserted 5' to exon 1, and a loxP-flanked neomycin selection cassete was inserted 3' to exon 1. Upon Cre-mediated recombination in ES cells, exon 1 and flanking intronic DNA were deleted, leaving a single loxP site in place of the deleted sequences. Western blot analysis of total protein lysates from MEFs showed that the endogenous CrkI and CrkII proteins were absent in homozyous mice. (J:111399)
