A targeting vector was designed to introduce the tyrosine 499 to phenylalanine substitution (Y499F) and a stop cassette into exon 13. An IRES-neomycin was included in the vector. RT-PCR confirmed the mutated sequence. Western blot detected reduced levels of both isoforms created by the locus. (J:111432)
