A targeting vector was designed to incorporate a 3-way stop codon at the 5' end of exon 21 and a neo cassette upstream of exon 21 that was subsequently removed via cre mediated recombination. Mutants demonstrated that the unique sequence of Nup96 was abolished and exon 20, which encodes the 6 kDa polypeptide shared by Nup98 and Nup96, was maintained so that expression of Nup98 remained uninterrupted.
