A vector was designed to replace 8 and 9 bp of the KI and KII sites (palindromic sequences, located in the intron between the Igkv and Igkj gene segments, involved in Igk cluster recombination), respectively. The sequence of KI was mutated from TCCACGCATGCTTGGA to TGTACGCTGCTGATGA and KII was mutated from TCCTCTTCAGTGAGGAG to AGCTCTTCTAGACTGA. A neomycin resistance gene cassette was deleted via cre-mediated recombination. (J:31727)
