Nucleotides -1404 through +61 of the spermatocyte-specific Pgk2 promoter region were joined to the cDNA encoding Cre recombinase followed by a rabbit globin polyadenylation signal. Expression of cre mRNA in testes was demonstrated by RT-PCR analysis. Double transgenic progeny of a cross of mice with this transgene to animals carrying a loxP-flanked EGFP reporter gene exhibit EGFP fluorescence in spermatocytes and spermatids, but not in spermatogonia or somatic cells of the testis.
