A targeting vector was designed to contain a 159-nt long human MYC sequence encoding the carboxy terminal part of the protein for insertion in place of the corresponding mouse sequence. The vector contained a neomycin resistance cassette with an RNA polymerase II promoter but without a polyA sequence immediately downstream of the MYC stop codon. The 3' UTR of the endogenous myc, including runs of Us, AREs and the two polyA sites was deleted and replaced by late SV40 3' UTR including a polyA site. (J:70610)
