A green fluorescent protein (GFP) cDNA was inserted into the first exon such that the endogenous promoter directs expression of GFP. An adjacent selectable marker was removed by cre-mediated recombination in ES cells prior to the production of chimeric mice. Expression of GFP was shown to mimic endogenous Il2 gene expression in both a temporal and spatial manner; induction experiments also showed that GFP expression was triggered by costimulatory signals similar to the endogenous gene.
